Fluorescence Imaging Analysis of Transient Trapping–microchip Micellar Electrokinetic Chromatography

This paper reports an fluorescence imaging analysis to verify the proposed mechanisms in transient trapping– microchip micellar electrokinetic chromatography (tr-trapping–MCMEKC). Fluorescence imaging revealed that a process of tr-trapping consisted of four migration stages with different apparent diffusion constants. In the first trapping stage, the diffusion of a focused sample band was suppressed on a boundary between the sample and partially injected micellar zones, while the sample bands slowly diffused in the micellar zone under the conventional sweeping condition. In the second releasing stage, it was clarified that the difference in the releasing time provided a highly effective separation compared to the conventional MCMEKC in spite of the shorter length of the micellar solution zone. In the third and fourth stages, the concentrated and separated samples migrated according to the principle of MEKC and zone electrophoresis, respectively. As a result, it was confirmed that the highly effective preconcentration and separation obtained in tr-trapping–MCMEKC was well explained by our proposed mechanism.
